I have only had my 360 for a few months now, so I would say Mass Effect, or the Orange Box.
Blue Dragon.
Oblivion. No question.
Gears of War. I have put the most time into that game than any other.
I've only had my 360 for about two-three weeks, so I don't have much experience with it. I'd go with Halo 3 just because the multiplayer is so damn enjoyable when you're playing with your friends.
It's probably for me Rock Band. Simply too amazing with 3 friends. However, GTA4 is simply an amazing work of art that's going to have me entranced for a long while.
Its between Assassins Creed and GTA IV in my opinion.

The Guitar Hero series, followed by Grand Theft Auto 4. Haven't really gotten my money's worth on next-gen yet. :(
